Abstract: The pyroligneous extract (PE) is a product of the pyrolysis process and is a mixture of organic compounds with relevant antimicrobial activity. In this work, we propose the optimization of the vacuum pyrolysis process of coconut endocarp and the best conditions to obtain PE was applied to other tree biomasses. Additionally, the characterization of the biomass, investigation of the chromatographic profile of the PE and evaluation of the effectiveness of PE against fungi that cause onychomycosis were performed. All evaluated biomasses showed similar thermal behavior, proximate and elemental characteristics. It was obtained a liquid yield (46.80–55.50%) by vaccum pyrolsis. Although the chromatographic profiles showed differences, they reproduced the same classes of compounds. The majority of identified compounds were phenolic compounds (relative area of 51.1–86.9%), followed by aldehydes (6.6–31.6%), ethers (1.6–4.8%) and ketones (0.43–9.9%). All the extracts evaluated showed antifungal activity against the tested microorganisms, showing inhibition halos with diameters ranging from 8 to 32 mm. The minimum inhibitory concentrations were 0.78–25.0 mg/mL for the coconut endocarp; 1.56–25 mg/mL for the dendê coconut endocarp; 1.56–25 mg/mL for the ouricuri endocarp, and 3.12–50.0 mg/mL for açaí seed. Furthermore, based on the novelty of the results obtained, a patent application for an antimycotic product of those extracts in the form of a solution, gel, hydrogel and aerosol was filed. Thus, PEs produced by sustainable routes are potential antifungal agent for the treatment of onchomycosis
