The patterns of linear dichroism (LD) and the dispersion of the birefringence (DB) were investigated in tendon collagen subjected to different histological procedures and staineâ with Toluidine Blue at pH 4.0 and 5.0. These characteristics showed to be variable when the LD sign, the displacement of the absorption peaks to different wavelengths, the occurrence of an inflection point in the DB curve, the extinction values and the ), of the troughs (or peaks) of the dichroic ratios are considered. The LD properties of the tendon stained at pH 4.0 seem to be a function o] the patterns of orientation of the AMPS (acid mucopolysaccharides) molecules whereas the Cotton effect-like phenomena demonstrated at pH 5.0 probably indicate the capacity of proteins and AMPS in ordering the dye molecules. Fixed material embedded in paraffin and [reezeâ unfixed material stained at pH 4.0 are suggested as controls for studies of the AMPS orientation and availability of their anionic groups in an oriented way.

Ce travail traite des caractéristiques du dichroisme linéaire (LD) et de Ia dispersion de Ia biréfringence (BD) du collagene tendineux soumis à divers traitements histologiques et coloré par le Bleu de Toluidine à pH 4,0 et 5,0. Ces caractéristiques se montrent variables en ce qui concerne le signe du LD, le déplacement des maxima dabsorption de diverses longueurs d'onde, l'apparition d'un point d'inflexion de Ia courbe DB (dispersion de Ia biréfringence), les valeurs d'extinction et du À des minima (ou des maxima) des indices du dichroísme (dll /d.lJ. Les propriétés du LD du tendon coloré à pH 4,0 semblent être fonction des types d'orientation des molécules AMPS (mucopolysaccharides acides ), alors que le phénomcne sernblable à un effet Cotton qui apparait à pH 5,0 traduit vraisemblablement Ia capacité des protéines et des AMPS d'ordonner les molécules du colorant. Pour servir de contrôle aux études de l'orientation des mucopolysaccharides acides et de Ia disposition de ses groupes anioniques de façon orientée, on a suggéré I'ernploi de matériel fixé et inclu en paraffine ou de matériel coupé au microtome de congélation et coloré à pH 4,0.
