Proteoglycan arrangement was investigated with polarizing and electron microscopes in the extracellular matrix of tendons of newborn rats. Based on linear dichroism data, the long axis of the AGAG molecules is supposed to be positioned predominantly parallel to the long axis of the collagen fibrils. Furthermore, a helical conformation is suggested for the AGAG molecules. Ultrastructural images of ruthenium red (RR) fixed material demonstrated dense globules connected among themse1ves and attached to the cell coat through fibrillary formations. These were seen to be attached to globules disposed on the same collagen fibril or connecting globules from different collagen fibrils. The fibrillary formations are assumed to represent proteoglycans, whereas the granules are supposed to be places where proteoglycans are attached to a linking protein (possibly a glycoprotein). Based on anisotropic data and ultrastructural images of RR-fixed material, a schematic model for the spatial arrangement of the components involved is proposed.

L'organisation des protéoglycanes [glycosaminoglycanes acides (GAGA)~omplexes protéiques] est étudiée par microscopie polarisée et microscopie électronique dans Ia matrice extracellulaire des tendons de rats nouveau-nés. En se basant sur des données de dichroísme linéaire, I'axe longitudinal des molécules GAGA est interprété comme étant situé en parallêle à l'axe longitudinal des fibres collagenes. En outre, une conformation en hélice est suggérée par les molécules GAGA. Les images électroniques obtenues à partir de matériaux traités au rouge de ruthénium (RR) démontrent Ia présence de globules denses reliés entre eux et rattachés à Ia membrane (cell-coat) cellulaire par des structures fibrillaires. Celles-ci peuvent rattacher soit des globules disposés sur une seule fibre, soit des globules appartenant à plusieurs fibres de collagêne. Les structures fibrillaires sont interprêtées comme représentant des protéoglycanes, alors que les globules correspondraient aux endroits ou les protéoglycanes sont rattachés à une protéine de support (une glycoprotéine, vraisemblablement). Les données d'anisotropie et d'images électroniques de matériaux fixés par RR suggêrent un modele schématique d'un arrangement spatial des divers éléments mis en évidence.
